چکیده

THE present work was undertaken as a preliminary step in the investigation of the experimental haemoglobinuria described by Callow and Hele [1927]. These workers found that p-bromoand p-chloro-phenylmercapturic acids, whether given by the mouth or subcutaneously, cause a transient haemoglobinuria in the dog. An attempt was made to find a small animal which would show a similar response to dosage with mercapturic acids. This attempt has not so far been successful; but in the course of the work certain interesting features of the sulphur metabolism of rats and rabbits were displayed. The possibility of mercapturic acid formation from iodobenzene and the effect of dosage with p-iodophenylmercapturic acid were studied in both these animals. The ability of the rabbit to synthesise p-bromophenylmercapturic acid has been shown by Abderhalden and Wertheimer [1925], and some evidence in favour of the occurrence of this synthesis in the rabbit has been brought forward by Rhode [1922], and by Shiple, Muldoon and Sherwin [1924] in the case of the pig. The evidence suggesting a synthesis of mercapturic acid by the rabbit, brought forward by Rhode, was very inconclusive, being founded solely upon the diminished synthesis of ethereal sulphate from monohalogen benzene derivatives when cystine was given simultaneously. Evidence in favour of the oxidation of mercapturic acid to ethereal sulphate has been adduced by Shiple, Muldoon and Sherwin [1924] for the case of the pig, and by Rose, Shiple and Sherwin [1924] for the case of the rabbit. On the other hand, previous work in this laboratory by Coombs and Hele [1927] indicated that this oxidation does not occur in the pig, and a criticism of the conclusions drawn by Sherwin and his co-workers is given in their paper. No case of haemoglobinuria following the administration of a mercapturic acid has been described, other than the observations of Callow and Hele quoted above; nor has haemoglobinuria been obtained, either in the rat or the rabbit, in the course of the present work.

Mercapturic acid precursors.

Although we have demonstrated previously that tissue slices are capable of carrying out a complete synthesis of p-iodophenylmercapturic acid (l), the intermediate compounds and the mechanisms involved in forming the link between the aromatic ring and cysteine sulfur have remained obscure. Various workers have suggested that a primary step is an attachment of the hydrocarbon to the sulfur of a p...

Total mercapturic acid synthesis by liver and kidney.

It has been known since 1879 that certain aromatic hydrocarbons and their halogen derivatives may be converted by the animal body to mercapturic acids as well as to various hydroxylated derivatives (1,2). The mercapturic acids generally have been isolated from the urine after high dosage of the hydrocarbon precursors. Synthesis of a mercapturic acid by isolated tissues has not been reported pre...

Pectic Acid Effects on Prolactin Secretion in GH3/B6 Rat Pituitary Cell Line

Background: Pectic acid extracted from plants increases the secretion of prolactin (PRL) when injected intravenously into ewes or fed to rats. Fragments of ewe hypophysis and lactating rabbit mammary gland incubated in vitro in the presence of pectic acid secreted more PRL and caseins compared to the controls. However, it is not known whether pectic acid directly stimulates PRL secretion in pi...

The Effects of Cholestasis and Cirrhosis on Gastric Acid and Pepsin Secretions in Rat: Involvement of Nitric Oxide

Objective(s) The liver has major role in the organism homeostasis, interactions with other systems, synthesis and metabolism of bile production, drug detoxification and hormone inactivation. Cholestasis can be defined as an impairment of the bile flow which can lead to hepatocytes necrosis and finally cirrhosis. Some studies reported a gastric acid secretion reduction in cirrhotic subjects, wh...

Inhibition of 5-Lipoxygenase inhibitor zileuton in high-fat diet-induced nonalcoholic fatty liver disease progression model

Objective(s): Arachidonic Acid/5-lipoxygenase (AA/5-LOX) pathway connects lipid metabolism and proinflammatory cytokine, which are both related to the development and progression of nonalcoholic fatty liver disease (NAFLD). Therefore, the present study was designed to investigate the role of AA/5-LOX pathway in progression of NAFLD, and the effect of zileuton, an inhibitor of 5-LOX, in this mod...
